Tyre puncture repair is a permanent fix for a hole in the tread: the tyre comes off the rim, the hole is cleaned out and a combined plug and patch is bonded from the inside. Only punctures in the central tread area can be repaired this way. Sidewall and shoulder damage cannot be repaired safely, and we will say so.

When can a tyre not be repaired?
When the damage is in the sidewall or shoulder, when the hole is too large, when the tyre was driven flat and the inside is scuffed, or when the tread is already at the limit.
Is a string plug from outside good enough?
It is a temporary fix. Nobody has looked inside the tyre, and a proper repair seals the liner as well as the hole.
How does this differ from your flat tyre fix?
The flat tyre fix is the roadside callout that gets you moving on the spare or with air. Puncture repair is the permanent fix to the tyre itself.
